Laboratory Test: Nicotine Air Testing in the Workplace (NIOSH 2544)

LCS Laboratory Inc. provides laboratory analysis for workplace nicotine exposure using the NIOSH Method 2544. The service is intended for industrial hygienists, EHS specialists, and consultants who need reliable data for exposure assessments and regulatory compliance.

With more than 18 years of experience in industrial hygiene laboratory testing, LCS analyzes nicotine samples collected in workplaces where tobacco is processed, handled, or stored.

Nicotine as an Occupational Hazard

Nicotine is a volatile organic compound with a boiling point of about 254 °C. It occurs naturally in tobacco leaves, typically at concentrations between 0.6 % and 3 % by weight, although the level varies depending on the plant variety and processing conditions.

Workers may be exposed to nicotine during:

  • harvesting and handling of tobacco leaves

  • tobacco processing and drying

  • manufacturing of tobacco products

  • storage and packaging operations

In addition to inhalation, nicotine can also be absorbed through the skin. For this reason, exposure assessments often consider both airborne concentrations and workplace handling practices.

In Ontario, the occupational exposure limit for nicotine is 0.5 mg/m³ (TWA) as vapour.

Sampling and Laboratory Analysis

Air sampling for nicotine is typically performed using XAD-2 sorbent tubes operated at approximately 1 L/min. Sampling times may extend up to 400 minutes, depending on the assessment strategy.

Samples are analyzed at LCS Laboratory using a modified version of NIOSH 2544:

  • sorbent extraction in the laboratory

  • analysis by gas chromatography with flame ionization detection (GC-FID)

  • capillary column separation

The typical analytical detection limit is about 1–3 µg per sample, which is sufficient for most occupational exposure assessments.

Laboratory Support for Industrial Hygienists

LCS Laboratory works with consultants and EHS teams across Canada. In addition to laboratory analysis, the lab can provide:

  • guidance on sampling strategy

  • air sampling pumps and media

  • basic training on sample collection

  • assistance interpreting laboratory reports
